How do I run OpenVPN on Vista?

To run OpenVPN-GUI on Vista:

run /bin/openvpn-gui.exe as administrator

add to config file:

route-method exe route-delay 2

As you found out, it works, but the ADD ROUTE command fails - the above changes will fix that (it’s a known bug in VISTA/OPENVPN-GUI).
